

Band 3 Antibody (Q1/156) is a mouse monoclonal IgG1 kappa light chain antibody that detects band 3 protein of human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Band 3, also known as AE1, is a crucial erythrocyte membrane glycoprotein that plays a significant role in maintaining cell structural integrity and facilitating chloride and bicarbonate ion exchange across the phospholipid bilayer. Band 3′s importance lies in dual functionality, mediated by a unique structure comprising approximately 900 amino acids organized into two distinct domains. The amino-terminal domain, referred to as the cytoplasmic domain of erythrocyte membrane band 3 (cdb3), serves as an essential attachment site for the erythrocyte cytoskeleton by binding to ankyrin, providing mechanical stability to the red blood cell. The carboxy-terminal, membrane-associated domain handles anion exchange transport, vital for maintaining acid-base balance in blood. Band 3 degradation can lead to formation of an aging antigen known as senescent cell antigen (SCA), expressed on older cells and signaling removal by the immune system, highlighting band 3′s role in cellular aging and homeostasis.
