ATP7A Antibody (D-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects ATP7A protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). ATP7A (D-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ATP7A, also known as Copper-transporting ATPase 1, is crucial for maintaining copper homeostasis in the body, functioning as a transmembrane copper-translocating P-type ATPase. ATP7A plays a significant role in systemic copper absorption in the gut and reabsorption in the kidney, which is vital for preventing copper deficiency and toxicity. ATP7A is primarily located in the trans-Golgi network and the basolateral membrane of polarized epithelial cells, such as those found in the kidney and intestine, where ATP7A facilitates the export of excess copper from cells into the bloodstream. While ATP7A is not typically expressed in most normal tissues, ATP7A is found in a significant fraction of various tumor types, where increased expression can confer resistance to chemotherapeutic agents like cisplatin and carboplatin. Mutations in the ATP7A gene are associated with Menkes disease, a severe disorder that leads to fatal outcomes in early childhood, while mutations in the ATP7B gene are linked to Wilson disease, which presents with neurological symptoms and liver damage.

Thank you for your question. This mouse monoclonal, ATP7A Antibody (D-9): sc-376467, is not raised against a peptide so no blocking peptide is available. The antibody is raised against a recombinant protein corresponding to amino acids 1-180 mapping within an N-terminal cytoplasmic domain of ATP7A of human origin. Because of the length of this protein, an antibody blocking or neutralization experiment would not be efficient. We would recommend performing alternative controls to evaluate the specificity of the antibody, for example using RNAi or CRISPR to silence or knockout the target gene.
