ATF-6α Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody designed to specifically target the activating transcription factor 6 alpha, also known as ATF-6α. ATF-6α monoclonal antibody (F-7) recognizes the epitope spanning amino acids 31-310 of the human ATF-6 protein and is validated for detecting ATF-6α in mouse, rat, and human samples using a variety of techniques including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). ATF-6α is a crucial member of the basic leucine zipper (bZIP) family of transcription factors that plays a vital role in the cellular response to endoplasmic reticulum (ER) stress. Under conditions of ER stress, ATF-6α undergoes proteolytic cleavage, allowing its active form to translocate to the nucleus where ATF-6α regulates the expression of ER chaperones and other genes essential for maintaining protein homeostasis and cell survival. This function is critical for understanding mechanisms related to protein folding diseases, cancer, and other stress-related pathologies. ATF-6α exists in cells as either an ATF-6β homodimer or an ATF-6α/β heterodimer, both of which bind to the nuclear transcription factor Y C (NF-YC) to induce the transcription of ER chaperone proteins. ATF-6α (F-7) antibody is available in both non-conjugated form and a range of conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatile options for diverse experimental applications.

I have the same question as olkr: does the antibody also recognise the cleaved form of ATF6? I am getting a strong band in ~50KDa, and was wondering if it is the cleaved form.

Thank you for your question. Anti-ATF-6α Antibody (F-7): sc-166659 was raised against amino acids 31-310 of ATF-6 of human origin and will recognize both full-length protein and fragments containing this amino acid sequence.

Does sc-166659 ATF6 antibody recognise both full-length and cleaved form of ATF6 (after using e.g thapsygargin) in Western blot application?

Can the flourochrome conjugated antibody distinguish between the 90kDa and 50kDa AFT6 expression by flow cytometry?

When should the TransCruz reagent sc-166659 X be used instead of the standard form sc-166659?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
