    Overview

    Aqp3 encodes aquaporin 3 (AQP3), an aquaglyceroporin plasma membrane channel that facilitates transmembrane transport of water, glycerol, and other small solutes, influencing cellular osmotic balance and metabolic substrate availability. In mouse tissues such as kidney, skin, and gastrointestinal epithelium, AQP3 contributes to epithelial barrier hydration, volume regulation, and glycerol-dependent energy metabolism, linking membrane transport to stress adaptation and proliferation. AQP3 activity can modulate redox-related signaling through effects on intracellular solute flux and is frequently studied in contexts of epithelial homeostasis, inflammation, and remodeling. Dysregulated AQP3 expression or localization has been associated with altered wound repair and inflammatory skin phenotypes, supporting its relevance for mechanistic studies of barrier function and disease-associated epithelial dysfunction.

    Aquaporin 3/AQP3 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Aqp3 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Aqp3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

    The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Aqp3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Aquaporin 3/AQP3 protein expression.

    This CRISPR knockout system enables efficient generation of Aqp3-deficient cell models for investigation of Aquaporin 3/AQP3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
