Ankyrin-1 Antibody (8C3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Ankyrin-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Ankyrin-1 antibody (8C3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Ankyrin-1 plays a crucial role in maintaining cellular structural integrity by linking integral membrane proteins to the cytoskeleton, which is essential for cellular stability and function. Ankyrin-1 is predominantly located in the plasma membrane of erythrocytes, cardiac and skeletal muscle, and neurons, where Ankyrin-1 facilitates the organization of membrane domains and anchoring of ion channels and transporters. This localization is vital for processes such as signal transduction and maintaining red blood cell shape, which is particularly important in preventing conditions like hereditary spherocytosis, a disorder characterized by red blood cell fragility. ANK1 gene encodes Ankyrin-1 in multiple isoforms, including a full-length version and several shorter variants, which may have distinct functional roles in different tissues.

Thank you for your question. Ankyrin-1 Antibody (8C3) is a mouse monoclonal antibody raised against Ankyrin-1 from erythrocytes of human origin. According to a publication in the literature (PMID: 11463472), the epitope for the 8C3 clone lies between amino acid residues 1 and 1012.

I am using Ankyrin-1 (8C3): sc-12733 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
