Amphiphysin I Antibody (G-4) is a mouse monoclonal IgG2b antibody that detects Amphiphysin I of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Amphiphysin I antibody (G-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Amphiphysin I is a crucial brain-enriched protein that plays a significant role in synaptic vesicle recycling, a process vital for neurotransmission. Amphiphysin I′s structure includes a membrane-bending BAR domain, a central Clathrin and adaptor binding domain, and a C-terminal SH3 domain, which collectively facilitate function in the formation and stabilization of membrane structures. In the brain, Amphiphysin I and II form heterodimers that interact with the Clathrin-associated GTPase Dynamin through their SH3 domains, a critical step that precedes the binding of Dynamin to Clathrin-coated pits, leading to vesicle budding. This interaction is essential for maintaining synaptic function and ensuring efficient neurotransmitter release. Additionally, in other tissues, Amphiphysin I may contribute to various membrane bending and curvature stabilization events, highlighting importance beyond the nervous system. The mammalian Amphiphysins share a similar overall structure, with a splice variant of Amphiphysin II that is highly expressed in muscle tissue, where Amphiphysin II is involved in the formation and stabilization of the T tubule network, further underscoring the diverse roles of Amphiphysin proteins in cellular dynamics.
