α-2M Antibody (H-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects the alpha-2M protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). α-2M monoclonal antibody (H-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Alpha-2-Macroglobulin (α-2M) is a homotetrameric serum protein composed of four identical subunits that dimerize through disulfide bonds, which is crucial for function as a pan-proteinase inhibitor. This unique structure allows α-2M to "bait" proteinases, leading to a conformational change that effectively traps and inhibits these enzymes, thereby regulating proteolytic activity in the body. Beyond serving as an inhibitor, α-2M also functions as a carrier protein for cytokines during inflammatory responses, binding to transforming growth factor β (TGFβ) and interleukin-6 (IL-6) to modulate activity and concentration in proximity to immune cells. Notably, mutations and deletions in the gene encoding α-2M have been linked to an increased risk of Alzheimer′s Disease, highlighting α-2M′s importance in the clearance of amyloid-beta, a key component in the pathology of this neurodegenerative condition.
