Alkaline Phosphatase Antibody (A-10) is a mouse monoclonal IgG2a kappa light chain antibody that detects Alkaline Phosphatase protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Alkaline Phosphatase antibody (A-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Alkaline phosphatases (AP) are crucial enzymes that play a significant role in dephosphorylation processes, which are vital for various physiological functions, including bone mineralization and cellular signaling. Placental alkaline phosphatase (PLAP), a tissue-specific isoform, is particularly important as PLAP is expressed in the placenta and found in the serum of pregnant women, serving as a potential biomarker for certain cancers, including ovarian and testicular tumors. PLAP involvement in cellular migration and transport of essential molecules, such as fatty acids and immunoglobulins, across the plasma membrane highlights PLAP importance in both reproductive biology and cancer research. The three tissue-specific alkaline phosphatases identified in humans—PLAP, germ cell alkaline phosphatase (GCAP), and intestinal alkaline phosphatase—exhibit 90-98% homology and are clustered on chromosome 2q, indicating a shared evolutionary origin and functional significance in various biological contexts.

Thank you for your question. Yes, this mouse monoclonal antibody, Alkaline Phosphatase (A-10): sc-271431, can be used in rat. We are aware of two publications citing use of the antibody in rat samples. These include PMID numbers: 25662160 and 26781793.

Rated 3 out of 5 by from Does not detect SEAP in western blotI tried to detect exogenously expressed SEAP (secreted alkaline phosphatase - a truncated version of PLAP) in HeLa cell lysate and media supernatant with this antibody in western blot (1:200 dilution). It didn't detect SEAP either in media or in cell lysate, even if the immunogen of this antibody should also be present in SEAP.

Rated 1 out of 5 by from A Blur of False Hope and Empty PromisesUsed a 1:250 dilution attempting to probe human osteosarcoma cells. The result was a faint blur at approximately the correct size, but by no means a well-define band. Attempted to incubate primary antibody again at 4 degrees overnight, and the band disappeared. No concentration of primary or secondary antibodies could resurrect it.
