AF-6 Antibody (B-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects AF-6 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). AF-6 (B-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AF-6 protein, also known as MLLT4 (myeloid/lymphoid or mixed-lineage leukemia translocated to 4), is a crucial 1,612 amino acid protein that plays a significant role in cell-cell adhesion and signaling. AF-6 contains two N-terminal Ras binding domains and a GLGF motif, which are essential for interaction with Ras proteins at peripheral cell junctions. AF-6 interacts with F-actin and Profilin, thereby influencing actin dynamics at adhesion complexes, which is vital for maintaining structural integrity of tissues. AF-6 coordinates recruitment of junction adhesion molecules (JAMs) to intercellular junctions through PDZ domain, highlighting importance in regulating cell polarity and junction integrity. Studies have shown that developing mice lacking AF-6 exhibit loss of neuroepithelial polarity, underscoring AF-6′s critical role in establishing and maintaining apical/basolateral asymmetry in epithelial tissues.

Can AF-6 (B-5): sc-74433 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, AF-6 (B-5): sc-74433 has been validated for use in IHC with paraffin-embedded sections.
