AE2 Antibody (D-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects AE2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). AE2 (D-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AE2 protein, also known as SLC4A2, plays a critical role in maintaining cellular homeostasis by regulating pH, chloride concentration, and cell volume through its function as a bicarbonate transporter. Located primarily in the canalicular membrane of hepatocytes and cholangiocytes, AE2 facilitates the exchange of chloride and bicarbonate ions, which is essential for the secretion of bicarbonate into bile. This bicarbonate secretion is vital for the generation of bile-salt independent flow, which is influenced by various factors such as hormones and biliary constituents. Disruption of AE2 function through gene silencing has been shown to significantly impair chloride/bicarbonate exchange, thereby affecting bile acid pool maintenance and the ability of secretin to stimulate bicarbonate-rich bile flow.
