Ada Antibody (ADA-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Ada protein of E. coli origin by western blotting (WB) and immunoprecipitation (IP). Anti-Ada antibody (ADA-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Ada protein plays a crucial role in the repair of alkylated guanine in DNA, which is vital for maintaining genomic integrity and preventing mutations that could lead to cell malfunction or disease. Ada protein is a monomeric, globular structure composed of 353 amino acids and features two distinct alkylacceptor domains that facilitate repair functions. Specifically, Ada protein can accept alkyl groups from the O-6 position of alkylguanine in DNA at cysteine residue 321 and from alkyl phosphotriester at cysteine residue 69, effectively inactivating the enzyme in the process. Ada protein is also capable of repairing O-4-methylthymine, showcasing versatility in DNA repair mechanisms. The N-terminal region of Ada protein is essential for the activation of transcription of genes such as ALKA, ALKB, and AIDB, highlighting regulatory role in cellular responses to DNA damage.
