Actin Antibody (8.S2.1) is a mouse monoclonal IgM kappa light chain antibody raised against monocytes and U-937 histolytic lymphoma of human origin, demonstrating reactivity with mouse, rat, human, and rabbit species. Actin plays a critical role in forming microfilaments within the cytoskeleton, existing in both globular (G-actin) and filamentous (F-actin) forms, which are essential for various cellular processes such as muscle contraction, cell motility, and maintenance of cell shape and integrity. Actin filaments can form both stable structures, like those in muscle sarcomeres, and dynamic structures, such as the leading edge of motile cells. While primarily localized in the cytoplasm, actin is also present in the nucleus, contributing to chromatin remodeling and gene transcription. The dynamics of actin are regulated by small GTPases like Rho, Rac, and Cdc42: Rho regulates the formation of stress fibers and focal adhesions; Rac controls actin accumulation at the plasma membrane leading to lamellipodia formation; and Cdc42 stimulates the formation of filopodia. Actin interacts with numerous proteins, including myosin for muscle contraction, tropomyosin and troponin for muscle regulation, and proteins like profilin and cofilin that regulate actin polymerization and depolymerization. These interactions are crucial for cell movement, division, and intracellular transport, underscoring the importance of actin in maintaining cellular function and structure. Anti-actin antibody (8.S2.1) is a valuable tool for researchers studying actin dynamics and related protein-protein interactions across various species.

Can Actin (8.S2.1): sc-70319 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: AbPolly
Thank you for your question. Yes, Actin (8.S2.1): sc-70319 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
