HSP 56 항체(329.1)는 마우스, 쥐 및 인간 유래의 HSP 56 단백질을 WB, IP 및 ELISA로 검출하는 IgG2a κ 마우스 단일 클론 HSP 56 항체입니다. HSP 56 항체(329.1)는 비접합 항 HSP 56 항체로 제공됩니다. HSP 56(FKBP4, FK506 결합 단백질 4, HBI, p52, FKBP52, FKBP59 및 PPIase라고도 함)은 면역필린 단백질 계열에 속하는 시스-트랜스 프롤릴 이성질화효소(시스-트랜스 프롤릴 이소머라제)입니다. 인간 HSP 56 유전자(FKBP4)는 여러 개의 폴리아데닐화 부위를 가지고 있으며 HSP 56 단백질은 인산화를 겪을 수 있습니다. HSP 56은 림프구에서 면역 조절 유전자 발현, 단백질 폴딩 및 트래피킹에 영향을 미칩니다. 또한 스테로이드 호르몬 핵 수용체의 공동 보호자 역할을 하여 표적 조직에서 적절한 호르몬 작용을 조절할 수 있습니다. 이 단백질은 일련의 테트라트리코펩타이드 반복(TPR) 도메인을 통해 피타노일-CoA 알파-하이드록실라제(PHYH) 및 HSP90과 결합할 수 있습니다. Why does one blot show no band for HSP56 in untransfected 293T cells the other blot does, and why don't you see more of it when over expressed? These data do not convince me that this antibody is actually staining for HSP 56

Thank you for your question. HSP proteins are generally pretty ubiquitously expressed so it is harder to show upregulation. The differences in signal observed between untransfected control 293T and HSP 56 transfected 293T could be a result of differences in protein loading, lysate preparations, exposure times, buffer quality, etc. There are many variables that can contribute to this observable difference. In particular, we used this antibody to detect protein levels in nuclear and cytosolic cell compartments in hippocampus and prefrontal cortex of mice and rats. In human samples we did not obtained protein bands detecting HSP56 levels. In general, we are not very satisfied with the obtained results, because Ab displayed low purity and high background in Western blot. Dilution to 1:200 for Western blot.
