세포카르틴 15 항체(LHK15)는 마우스, 랫트, 인간에서 유래된 세포카르틴 15 단백질을 웨스턴 블롯(WB), 면역침전법(IP), 면역형광법(IF), 면역조직화학법(IHC)을 통해 검출하는 마우스 단일클론 IgG2a 카파 경쇄 항체입니다. 항-세포카르틴 15 항체(LHK15)는 비접합 형태와 아가로스, 호스래디시 퍼옥시다아제(HRP), 피코에리트린(PE), 플루오레세인 이소티오시아네이트(FITC), 그리고 여러 가지 알렉사 플루오르® 접합체를 포함한 다양한 접합체 형태로 제공됩니다. 세포각질 15(CK15, K15, K1CO, keratin 15, type I cytoskeletal 15)는 상피세포, 특히 모낭의 기계적 완전성을 유지하는 데 중요한 역할을 하는 중간 필라멘트 단백질입니다. 세포카르틴 15는 주로 모낭 돌출부에 위치하며, 이 부위에서 줄기세포의 특정 표지자 역할을 함으로써 모낭 생물학과 재생에 있어 세포카르틴 15의 중요성을 강조합니다. 세포카르틴 15 발현은 기저세포암과 상피모세포종(trichoepithelioma)을 구분하는 데 중요한 역할을 하는데, 두 질환 모두 진단을 복잡하게 만들 수 있는 중복되는 형태학적 특징을 나타내기 때문입니다. 모발 각질(hair keratin)의 부재와 함께, 세포각질15와 세포각질19의 발현 패턴은 이러한 종양의 분화 상태에 대한 귀중한 통찰을 제공하여, 세포각질15 단일클론항체(LHK15)를 피부병리학 연구자와 임상의에게 필수적인 도구로 만들어 줍니다.

Can Cytokeratin 15 (LHK15): sc-47697 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, Cytokeratin 15 (LHK15): sc-47697 monoclonal antibody has been validated for use in IHC with paraffin-embedded sections. Rated 5 out of 5 by from works very well in histology alone or in coIFSerial sections from human scalp biopsies and mouse ears embedded in OCT or human scalp biopsies embedded in formalin‐fixed paraffin were processed for immunofluorescence as follow: Frozen sections were fixed with neutral buffered 4% paraformaldehyde (PFA) for 15 min at room temperature (RT) and washed three times with phosphate‐buffered saline (PBS). Paraffin sections were processed for their deparaffinization and antigen retrieval. Primary antibody specific for Keratin 15 at 1:400 dilution was incubated overnight (O/N) at 4°C. Sections were then washed three times with PBS and incubated with secondary antibody conjugated to the appropriate fluorophore(s) (for example, Alexa Fluor 647 dilution 1:200) for 1 h at RT. Sections were then washed three times with PBS, counterstained with DAPI, mounted with IF-grate antifade mounting medium and analyzed by fluorescence or confocal microscopy.
