TMEM125 CRISPR/Cas9 KO Plasmid (h) | sc-414318 | 20 µg | $397.00 |
TMEM125 encodes a multi-pass transmembrane protein with emerging roles in membrane organization and intracellular trafficking, processes that influence receptor turnover and compartmentalized signaling. Its expression has been reported in multiple tissues, and transcript-level dysregulation has been observed in several disease datasets, suggesting potential relevance to cellular homeostasis under stress. Although mechanistic annotation remains limited, TMEM125 is frequently evaluated in functional genomics efforts focused on membrane-proximal pathways, organelle dynamics, and proteostasis. These features make TMEM125 a useful target for dissecting how membrane proteins shape signaling networks and cell-state transitions.
TMEM125 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TMEM125 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TMEM125 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TMEM125 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TMEM125 protein expression.
This CRISPR knockout system enables efficient generation of TMEM125-deficient cell models for investigation of TMEM125 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
