SPT5 Antibody (D-10) is a mouse monoclonal IgG1 antibody that detects SPT5 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SPT5, also known as DSIF p160, plays a crucial role in the regulation of transcriptional elongation, particularly in RNA polymerase II activity. SPT5 is essential for forming the DRB-sensitivity-inducing factor (DSIF) complex, which forms when SPT5 binds to SPT4. The DSIF complex is vital for modulating the transcriptional process, as SPT5 interacts directly with RNA polymerase II to facilitate elongation. Notably, SPT5 undergoes post-translational modifications, particularly phosphorylation, which significantly influences function. In mitotic HeLa cells, SPT5 exhibits slower migration on SDS-PAGE compared to its interphase counterpart due to increased phosphorylation levels, highlighting the importance of these modifications in regulating activity. The C-terminal CTR1 domain of SPT5 is specifically targeted for phosphorylation by the P-TEFb complex, a modification that is critical for SPT5′s role as a transcriptional elongation regulator.
