Podocalyxin-like 1 Antibody (4F10) is a mouse monoclonal IgM kappa light chain antibody raised against human recombinant podocalyxin, effectively detecting podocalyxin-like protein 1 (PODXL) of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). PODXL, also known as PCLP1 or PODXL1, is a transmembrane sialomucin characterized by a heavily glycosylated extracellular domain, a single-pass transmembrane region, and a cytoplasmic tail containing multiple phosphorylation sites. The structure of PODXL is crucial for maintaining cell polarity and contributing to the glomerular filtration barrier in kidney podocytes; extensive glycosylation and negative charge help repel adjacent foot processes, preventing their fusion. Structurally related to CD34, PODXL acts as an anti-adhesive molecule, regulating cell-cell and cell-matrix interactions important in vascular development and hematopoiesis. PODXL is expressed in various epithelial tissues, including breast, pancreas, and prostate, in addition to endothelial cells and hemangioblasts. PODXL is involved in cancer progression and metastasis, particularly in breast and prostate cancers, highlighting relevance in oncological research. Anti-PODXL antibody (4F10) recognizes the extracellular domain of PODXL, serving as a valuable tool for studying roles in various biological processes, including inflammation, where PODXL mediates leukocyte adhesion to endothelial cells. PODXL interacts with the actin cytoskeleton via ezrin, which is essential for maintaining cell shape and polarity, and serves as a ligand for L-selectin, facilitating leukocyte rolling on endothelial cells during inflammatory response. PODXL monoclonal antibody (4F10) in non-conjugated form allows versatility in experimental applications, aiding researchers in exploring structural and functional relationships of PODXL in normal physiology and disease states.

How much protein do you recommend loading on the gel for Western blot detection using Podocalyxin-like 1 (4F10): sc-23903 monoclonal antibody?

Asked by: jenniferc
We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting
