PHF3 (PHD finger protein 3) is a nuclear transcriptional regulator that associates with RNA polymerase II and contributes to control of transcription elongation and co-transcriptional RNA processing. Through its PHD finger domains and interactions with elongation machinery, PHF3 influences gene expression programs linked to cell-state maintenance and stimulus-responsive transcription. Perturbation of PHF3 activity has been connected to altered transcriptional fidelity and dysregulated growth-control networks, making it relevant for studying mechanisms that couple chromatin-associated factors to RNA synthesis. PHF3 is therefore of interest in research areas spanning gene regulation, differentiation, and disease-associated transcriptional rewiring.
PHF3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the PHF3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the PHF3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the PHF3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish PHF3 protein expression.
This CRISPR knockout system enables efficient generation of PHF3-deficient cell models for investigation of PHF3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
