SEE ALSO...

D5DR Antibody (E-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects D5DR protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-D5DR antibody (E-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. D5DR protein, also known as dopamine receptor D5, DR1B, or DRD1L2, is a G protein-coupled receptor belonging to the D1-like family, which plays a crucial role in stimulating adenylyl cyclase and exhibits a tenfold higher affinity for dopamine compared to the D1 subtype. D5DR is predominantly located in neurons within the limbic regions of the brain, with mRNA being most abundant in specific cortical areas (layers II, IV, and VI), the dentate gyrus, and various hippocampal subfields, while showing lower expression levels in the striatum. D5DR′s strategic location is vital for modulating neurotransmission and influencing behaviors associated with motivation, reward, and cognition. Five major types of dopamine receptors exist, all G protein-coupled metabotropic receptors that can exert either excitatory or inhibitory effects on post-synaptic neurons. D1 (D1A, D1C, D1D) and D5 (D1B) receptors are part of the D1-like receptor family, which typically promotes excitatory signaling through increases in cyclic adenosine monophosphate (cAMP), while D2, D3, and D4 receptors belong to the D2-like family, which generally inhibits cAMP production. Research has shown a significant age-related decline in dopamine receptor mRNAs, particularly in the hippocampus and entorhinal cortex, highlighting D5DR′s relevance in neurobiological studies and potential therapeutic targets for age-related cognitive decline.

Can D5DR (E-12): sc-376088 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jerojero
Thank you for your question. Yes, D5DR (E-12): sc-376088 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
