CD3-ζ Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD3-ζ in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). CD3-ζ (D-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD3-ζ protein plays a crucial role in T cell activation, as CD3-ζ is part of the T cell receptor (TCR) complex that transmits signals upon recognition of foreign antigens. This signaling is essential for the immune response, as CD3-ζ leads to the activation of T cells, which are vital for adaptive immunity. The CD3 complex, which includes CD3-ζ, consists of five invariant polypeptide chains that form three distinct dimers, facilitating the transmission of activation signals. The ζ chain contains three immunoreceptor tyrosine-based activation motifs (ITAMs), which are critical for recruiting protein kinases like ZAP-70 and Syk, thereby amplifying the signaling cascade that results in T cell activation. The structural integrity and phosphorylation state of these motifs are vital for effective immune responses, making CD3-ζ a key player in immunological signaling pathways.

Thank you for your question. Yes, CD3-ζ (D-4): sc-374281 has been validated for use in IHC with paraffin-embedded sections.
