caspase-3 Antibody (3C119) is a mouse monoclonal IgG2a kappa light chain antibody designed to detect both active caspase-3 and its inactive precursor, procaspase-3, across mouse, rat, and human species. Caspase-3, also known as apopain, SCA-1, Yama, and CPP32, is a crucial aspartate-specific cysteine protease within the ICE subfamily, playing a central role in the execution phase of apoptosis. The structure of caspase-3 consists of large (p17) and small (p11) subunits that form an active heterotetramer upon post-translational cleavage, a modification essential for proteolytic activity and regulation of programmed cell death. This post-translational processing transforms procaspase-3 from an inactive zymogen into an active enzyme capable of cleaving key substrates such as PARP, thereby facilitating the orderly disassembly of cellular components during apoptosis. Additionally, activated caspase-3 can further cleave and activate other caspases, including caspase-6, -7, and -9, amplifying the apoptotic signal. The precise control of caspase-3 activation through these post-translational modifications is vital for maintaining cellular homeostasis and preventing uncontrolled cell death, which is implicated in various diseases including cancer, neurodegenerative disorders, and immune system dysfunctions.
