rhodopsin Antibody (1D4) is a high quality monoclonal rhodopsin antibody (also designated rhodopsin antibody) suitable for the detection of the rhodopsin protein of mouse, rat and human origin. rhodopsin Antibody (1D4) is available as both the non-conjugated anti-rhodopsin antibody form, as well as multiple conjugated forms of anti-rhodopsin antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. G protein-coupled receptors (GPCRs), which are characterized as containing seven transmembrane α helices, elicit G protein-mediated signaling cascades in response to a variety of stimuli. The opsin subfamily, which represents approximately 90 percent of all GPCRs, is comprised of photoreceptors that are activated by light, which include the red-, green- and blue-sensitive opsins and rhodopsin. The opsin subfamily consists of an apoprotein covalently linked to 11-cis-retinal, which undergoes isomerization upon the absorption of photons. This isomerization leads to a conformational change of the protein which results in the activation of hundreds of G proteins. Specifically, rhodopsin exhibits a maximal absorption at 495 nm and mediates vision in dim light. Mutations in the rhodopsin gene lead to retinitis pigmentosa, which can be inherited as an autosomal dominant, an autosomal recessive or an X-linked recessive disorder.

Would this antibody recognize the 1D4 tag in fusion protein in western blot?

Thank you for your question. This is a mouse monoclonal antibody that is recommended for detection of the protein rhodopsin. The clone name for this antibody is “1D4”, which is also known as the “1D4” epitope of rhodopsin. We have not tested this antibody for detection of any 1D4 fusion proteins.

Can this mouse monoclonal antibody be used in IF or IHC on mouse tissue without background or non-specific staining issues?

Thank you for your question. Yes, it can be used successfully for this application. We recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody rhodopsin (1D4): sc-57432 is currently available conjugated to HRP, PE, FITC, AF488 and AF647.

Visualizing Endogenous Rhodopsin in RPE CellsUseful antibody for IF labeling of endogenous rhodopsin in RPE cells. Cells permeabilized for 30min in PBStween then blocked in 5%BSA. Antibody used at 1:200 overnight at 4 degrees.

Antibody for IFI used this antibody on OCT embedded frozen retinal sections (mouse). Blocked for 1 hour with 10% goat serum buffer. Incubated with primary antibody overnight at the a concentration of 1:100. Incubated with secondary antibody (alexa fluor 568 anti-mouse) at a concentration of 1:1000. Staining was fairly robust but not completely consistent between all samples.
