PML Antibody (PG-M3) is a mouse monoclonal IgG1 kappa light chain antibody that detects PML protein of human origin through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry (IHC). PML (PG-M3) monoclonal antibody is offered in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. PML functions as a zinc finger transcription factor and is crucial for regulating gene expression involved in cell differentiation and apoptosis. PML′s role in these processes is particularly significant in acute promyelocytic leukemia, where PML gene undergoes a chromosomal translocation with the retinoic acid receptor α (RARα) gene, resulting in the formation of the PML/RARα chimeric protein. This aberrant protein disrupts normal myeloid differentiation by recruiting the nuclear co-repressor-histone deacetylase complex, which is essential for retinoic acid-dependent differentiation, and also induces gene hypermethylation, leading to the silencing of critical genes and promoting leukemogenesis. PML′s ability to influence these pathways underscores PML′s importance in both normal cellular function and disease states, making PML (PG-M3) antibody a valuable tool for researchers studying these mechanisms.
