Pirh2 Antibody (E-10) is a mouse monoclonal IgG2b antibody that detects Pirh2 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Pirh2, also known as Androgen receptor N-terminal-interacting protein (ARNIP), ZN363, or CHIMP, plays a critical role in cellular regulation by functioning as a p53-induced ubiquitin-protein ligase, which promotes the degradation of the tumor suppressor protein p53. The importance of this interaction lies in the fact that p53 is a key regulator of the cell cycle and apoptosis, and Pirh2-mediated degradation can lead to oncogenic processes, as the loss of p53 function is associated with the development of malignant tumors. The gene encoding Pirh2 is located on chromosome 4q21.1, and Pirh2 transcription is intricately regulated by p53 itself, creating a feedback loop that modulates cellular responses to stress and DNA damage. Notably, Pirh2 expression inversely correlates with p53 levels; when Pirh2 is overexpressed, p53 levels decrease, and conversely, a reduction in Pirh2 expression leads to an increase in p53.
