p-EGFR Antibody (15A2) is a mouse monoclonal IgG1 antibody that detects phosphorylated Epidermal Growth Factor Receptor (p-EGFR), of mouse, rat, human, and canine origin by western blotting (WB) and immunoprecipitation (IP). p-EGFR monoclonal antibody (15A2) is available as the non-conjugated anti-p-EGFR antibody form. The Epidermal Growth Factor Receptor (EGFR) plays a crucial role in regulating cell growth, survival, and differentiation through interactions with epidermal growth factors. Upon binding of ligands such as EGF or TGFα, EGFR undergoes a conformational change that activates intrinsic tyrosine kinase activity, leading to autophosphorylation at key tyrosine residues, including Tyr 1068, Tyr 1092, and Tyr 1173. These phosphorylation sites serve as docking platforms for signaling proteins containing SH2 or PTB domains, ensuring precise recruitment and activation of downstream effectors. Autophosphorylation also induces structural changes in EGFR′s intracellular domain, enhancing interaction with multiple signaling proteins simultaneously, amplifying signal transduction. These phosphorylation events are essential for recruiting downstream signaling proteins, such as GRB2, which facilitate activation of the Ras signaling pathway, a critical pathway for cell proliferation and survival. p-EGFR′s ability to mediate these signaling cascades underscores importance in various physiological processes and potential role in cancer progression, making p-EGFR a valuable target for therapeutic intervention and research. anti-p-EGFR antibody (15A2) provides researchers with a reliable tool to study EGFR phosphorylation and role in key signaling pathways, advancing understanding of function in normal physiology and oncogenesis.

The data sheet says that Tyr1068 is also designated as Tyr1092. The protein reference NP_005219.2 shows me a Tyr at 1069 and at 1092. So which one does this antibody detect now?

Asked by: ToxVogel
Thank you for your question. These two phosphorylation sites are actually the same site, it's the numeration that varies for this protein. The site this antibody detects is in literature often referred to as Tyr 1068, but according to the swiss-prot accession#P00533, it is Tyr1092.

I have purchase this antibody: "https://www.scbt.com/scbt/product/p-egfr-antibody-15a2"; but there are always two bands shown. one is near 170, the other is around 130. Is that right?
