


BRDG1 Antibody (G-5) is a mouse monoclonal IgG2a antibody that detects BCR downstream signaling-1 (BRDG1) in mouse samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). BRDG1 plays a crucial role in enhancing the activity of Tec kinases, which are vital for mediating cellular responses downstream of B cell antigen receptor activation, as well as other signaling pathways involving cytokine receptors and integrins. BRDG1 functions as a docking protein, characterized by a proline-rich pleckstrin homology (PH) domain and multiple tyrosine phosphorylation sites, allowing interaction with SH2 domain-containing effector proteins. The phosphorylation of BRDG1 by Tec family nonreceptor protein tyrosine kinases, such as Tec and Pyk2, is essential for signal transduction, as BRDG1 facilitates the recruitment of downstream signaling molecules that amplify the cellular response. The human BRDG1 protein is encoded by a gene located on chromosome 4q13.1 and consists of 295 amino acids, with transcripts being particularly abundant in the Ramos human B cell line. The mouse ortholog, stem cell adaptor protein 1 (STAP-1), shares a remarkable 83% identity with BRDG1, highlighting the evolutionary conservation of this signaling pathway.
