Myosin IXa is a protein encoded by the MYO9A gene in humans. It belongs to the myosin superfamily, a diverse group of motor proteins known for their roles in muscle contraction and a wide variety of other cellular processes. Myosin IXa is considered an unconventional myosin due to its unique structure and function compared to the more well-known muscle myosins.Structurally, Myosin IXa contains a motor domain that can bind ATP and actin, a regulatory domain, and a tail domain. Uniquely, the tail domain has a RhoGAP domain, which can negatively regulate small GTPases, particularly Rho-type GTPases. These GTPases control a variety of cellular processes, including cytoskeletal dynamics and cell migration.
Functionally, Myosin IXa is involved in actin-based intracellular transport and cell motility. By interacting with the actin cytoskeleton, it plays a critical role in maintaining cell shape and polarity. It also contributes to intracellular signal transduction through its RhoGAP activity.Moreover, Myosin IXa is implicated in the regulation of cell adhesion, contributing to processes such as wound healing and immune response. It also plays a role in the development of neuronal cells and may be involved in the formation of dendritic spines, which are small protrusions on neurons involved in transmitting synaptic signals.
