L'anticorps Smo (E-5) est un anticorps monoclonal de souris IgG2a κ Smo (également appelé anticorps Smoothened) qui détecte la protéine Smo chez la souris, le rat et l'homme par WB, IP, IF et ELISA. L'anticorps Smo (E-5) est disponible sous forme d'anticorps anti-Smo non conjugué, ainsi que sous plusieurs formes conjuguées d'anticorps anti-Smo, y compris agarose, HRP, PE, FITC et plusieurs conjugués Alexa Fluor®. La surexpression de la protéine Wnt-1 ou de la protéine GLI est à l'origine du cancer, mais la base moléculaire de cette transformation était mal comprise. Les protéines Wnt-1 et GLI ont été placées dans une cascade de signalisation en aval des homologues mammaliens des protéines Hedgehog et Patched de la drosophile. Le gène de polarité segmentaire hedgehog (hh) de la drosophile code pour une protéine sécrétée qui semble jouer un rôle dans la structuration de l'embryon et du disque imaginal. Le gène ptc, également identifié comme un gène de polarité segmentaire de la drosophile, code pour la protéine transmembranaire patched, dont l'expression est précisément régulée au cours du développement embryonnaire. Il a été démontré que Hedgehog renforce l'expression des protéines de la famille Wnt par le biais d'une cascade de signalisation impliquant les facteurs de transcription GLI, tandis que patched fonctionne comme un répulseur s'opposant aux effets de Hedgehog. Smoothened (Smo), un récepteur à sept transmembranes, est complexé avec patched dans de nombreux tissus et est considéré comme un composant essentiel de la voie de signalisation Hh.

When I use this antibody in Western blot experiments, I always observe multiple bands. I wonder if this could be related to my denaturation method. I usually add 5× loading buffer to the protein lysate and incubate it at 37°C for 30 minutes. Do you have

Hello, my lab works with S. purpuratus sea urchins. Could you please check if the antibody will be compatible with our species? This is the NCBI reference sequence for the Smo protein in urchins: XP_030830155.1

Has this antibody been tested in Xenopus, either WB or IF?

Thank you for your question. Anti-Smo Antibody (E-5) has only been tested for detection of Smo of mouse, rat and human origin by WB, IP, IF and ELISA. It has not been tested in samples from Xenopus.

Thank you for your question. This antibody was raised against amino acids 488-787 of Smo of human origin, which spans both cytoplasmic and extracellular regions. The exact epitope has not been epitope mapped. If you have any further questions, please contact Technical Service.

Rated 3 de 5 de par good for western blot analysisWe used this antibody to detect Smo and got a good result. Fresh liver samples were homogenized in RIPA buffer, then 40ug of total protein were subjected to 10% gel for SDS-PAGE and transferred onto a PVDF membrane. After blocking with 5% skim milk in 1×TBS-T, membrane were incubated overnight at 4℃ with Smo antibodies(1:1000).And then, it were incubated with anti- mouse IgG(1:2000) at room temperature and detected with ECL kit.
