NEDD8 Antibody (H-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects NEDD8 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-NEDD8 antibody (H-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. NEDD8, a ubiquitin (Ub)-like molecule, shares 80% homology with ubiquitin and plays a crucial role in regulating protein degradation pathways. NEDD8 is primarily localized in the nucleus and is highly expressed in adult heart and skeletal muscle, which is significant for maintaining cellular homeostasis and muscle function. NEDD8 is activated by the E1 ubiquitin-activating enzyme UBA2, forming isopeptide linkages that facilitate attachment to target proteins. NEDD8 is covalently linked to an E3 ubiquitin ligase through the E2 conjugating enzyme Ubc12, preferentially associating with the CUL-2 protein in the E3 ligase complex CBCVHL. This interaction is vital for the regulation of various cellular processes, including cell cycle progression and signal transduction. In vitro studies have shown that NEDD8 tetramers can bind to the 26S proteasome, suggesting a role in modulating proteolysis similar to other ubiquitin-mediated pathways.
