L'anticorps Histone H3 (1G1) est un anticorps IgG monoclonal de souris qui détecte l'Histone H3 dans des échantillons de souris, de rats et d'humains par western blotting (WB) et immunofluorescence (IF). L'anticorps anti-Histone H3 (1G1) est disponible sous forme non conjuguée. L'histone H3 joue un rôle crucial dans l'organisation structurelle de la chromatine, essentielle pour l'empaquetage de l'ADN dans le noyau. L'histone H3 forme une partie centrale de la structure octamérique du nucléosome qui enveloppe l'ADN, régulant l'expression des gènes et la réplication de l'ADN. L'histone H3 maintient la stabilité du génome et facilite les processus de transcription et de réparation. L'histone H3 subit diverses modifications post-traductionnelles, notamment la méthylation et l'acétylation, qui influencent les interactions entre l'ADN et les protéines, ainsi que la structure et la fonction de la chromatine. Ces modifications régulent l'expression des gènes et ont un impact sur la différenciation cellulaire et la réponse environnementale. Les capacités de détection de l'anticorps anti-histone H3 (1G1) rendent ce réactif précieux pour l'étude de la dynamique de la chromatine et de la régulation épigénétique dans divers contextes biologiques.

I performed ICC with this antibody in mitotic cells, but I couldn't detect histone H3 signal co-localized with the DAPI signal. Could you help me solve this issue?

Just received a free sample, which dilution would you recommend for nuclear extracts WB?

Thank you for your question. Dilution recommendations can be found in the product's datasheet, which is available right here on the product page. For western blots we recommend a starting dilution of 1:200, and a dilution range between 1:100-1:1000. When working with nuclear extracts, an amount of protein between 10–20 µg is appropriate.

Rated 5 de 5 de par H3 Ab for Western BlottingAsked for a sample before purchasing. Worked straight away on CRC cell lines HCT116 and DLD1 (drug treated and untreated) when loading 20 ug of whole protein lysates and using 1:500 dilution in 5% BSA/TBST. Would definitely recommend.

Too strong signal with the dilution you recommend. Conditions need to be better set. Apparent MW: about 15kDa. Expected MW: 15kDa. • 40ug/lane cell lysate • Transfer 1h30 100V, PVDF (0.2 µm) • Blocking milk 4% in 0.1% Tween -PBS 1h RT • Anti-HISTONE H3 (1G1)(sc-517576) 1:200 O/N 4°C in blocking solution • Anti-mouse HRP 1:1000 1h RT in blocking solution • Developed with ECL Plus
