Hephaestin Antibody (C-7) is a mouse monoclonal IgG2b kappa light chain antibody that detects Hephaestin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Hephaestin (C-7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Hephaestin is a single-pass type I membrane protein belonging to the multicopper oxidase family, and plays a vital role as a copper-dependent ferroxidase, facilitating iron export from intestinal enterocytes into the bloodstream. This function is crucial for maintaining iron homeostasis, ensuring iron availability for essential biological processes, including hemoglobin synthesis and cellular respiration. Hephaestin operates in conjunction with ferroportin 1 to mediate iron transport across the basolateral membrane of enterocytes, linking iron and copper metabolism in mammals. Notably, copper deficiency can lead to decreased Hephaestin levels, resulting in impaired iron absorption and subsequent anemia. Hephaestin can bind up to six copper ions per monomer and is regulated by the homeobox transcription factor CDX2, with increased iron levels stimulating CDX2 expression and, consequently, Hephaestin production. While primarily expressed in the intestine, Hephaestin is also found in the colon, breast tissue, bone trabecular cells, and fibroblasts, highlighting diverse roles in various tissues.

We recommend using one of our our exclusive Mouse IgG Binding Proteins conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
