L'anticorps anti-E-cadhérine (67A4) est un anticorps monoclonal IgG1 à chaîne légère kappa de souris qui détecte la E-cadhérine humaine par western blotting (WB), immunoprécipitation (IP), immunofluorescence (IF) et cytométrie de flux (FCM). L'anticorps anti-E-cadhérine (67A4) est disponible sous forme non conjuguée et sous diverses formes conjuguées, notamment agarose, peroxydase de raifort (HRP), phycoérythrine (PE), isothiocyanate de fluorescéine (FITC) et plusieurs conjugués Alexa Fluor®. La E-cadhérine, membre de la famille des cadhérines, joue un rôle crucial dans l'adhésion cellule-cellule, essentielle au maintien de l'intégrité des tissus et à la facilitation des processus morphogénétiques au cours du développement. La E-cadhérine se localise principalement à la membrane cellulaire, formant des jonctions d'adhérence qui relient les cellules adjacentes et contribuent à la stabilité structurelle des tissus. Cette localisation membranaire permet à la E-cadhérine d'assurer une forte adhésion intercellulaire, nécessaire à la formation de la couche épithéliale et à la prévention des comportements invasifs des cellules cancéreuses. L'interaction de la E-cadhérine avec les protéines cytoplasmiques, telles que la β-caténine, est essentielle pour les voies de signalisation qui régulent la prolifération et la différenciation cellulaires, soulignant son importance à la fois dans la physiologie normale et dans les états pathologiques.

Rated 1 de 5 de par Anticorpo non funzionantePrimary antibodies E-Cadherin (67A4) sc-21791 was used diluted 1:500 according to the manufacturer’s recommendations (for Western Blotting dilution range 1:100-1:1000), in Milk 5%. Membrane of PVDF has been incubated overnight at 4°C and then it has been probed for 1h at RT with Santa Cruz secondary antibodies diluted 1:5000 in Milk 1%. We have tested this antibody on two different sets of samples: in six cancer cell lines and in human activated PBMCs. Moreover, even if we tested the antibody at higher concentration, we only obtained a higher background but not an increase in the specific protein signal; therefore we show only the blots obtained after the incubation with antibody prepared as described above. In conclusion: • E-Cadherin did not work at all, neither after a long exposure. We do not have tools to verify the specificity for this antibody too. All the membranes have been probed with a normalizer and each gel has been stained with Coomassie Brillant Blue, shown on the right side of each panel, in order to demonstrate that the samples ran well and that the proteins are present on the SDS-PAGE.
