CD73 Antibody (2B6) is a mouse monoclonal IgG2b antibody that detects CD73 of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-CD73 antibody (2B6) is available as the non-conjugated form. CD73, also known as 5′-nucleotidase, E5NT, NT, NT5, NTE, eN, and eNT, is a glycosyl-phosphatidylinositol (GPI)-anchored adhesion protein that plays a crucial role in the regulation of extracellular nucleotide metabolism by catalyzing the dephosphorylation of purine and pyrimidine nucleotides into their corresponding bioactive nucleosides. This enzymatic activity is vital for maintaining immune homeostasis and modulating inflammatory responses, as CD73 influences the availability of nucleosides that can act on various receptors, thereby affecting cell signaling pathways. CD73 is structurally characterized as a dimer of two identical subunits, which relies on its GPI anchor for attachment to the external face of the plasma membrane. CD73′s location and anchoring mechanism are significant because these features facilitate interaction with other cell surface proteins and enable mediation of co-stimulatory signals during T cell activation. CD73 exhibits diverse biological functions through differential regulation in various cell types, including endothelial cells, subpopulations of B and T cells, germinal center follicular dendritic cells, and thymic medullary reticular fibroblasts. Notably, CD73 neutralization by IgG can disrupt lymphocyte adhesion to endothelial cells and inhibit the aggregation of germinal center B cells and follicular dendritic cells, highlighting CD73′s importance in immune cell interactions. Furthermore, targeting lymphocyte CD73 can lead to shedding and subsequent tyrosine phosphorylation of associated proteins, underscoring CD73′s multifaceted role in immune regulation.
