Lipopolysaccharide (LPS) elicits the secretion of mediators and cytokines produced by activated macrophages and monocytes. CD14 is a glycosylphosphatidylinositol (GPI)-anchored protein found on the surfaces of monocytes and polymorphonuclear leukocytes. CD14 functions as a receptor for LPS, resulting in the secretion of various proteins. An important component in the LPS activation of monocytes through the CD14 receptor is the "adapter molecule," lipopolysaccharide binding protein (LBP). There are two forms of CD14, a membrane-associated form (mCD14), and a soluble form (sCD14). mCD14 responds to LPS alone and facilitates the secretion of proteins, while cells not expressing mCD14 fail to respond to LPS. The cells that lack mCD14 respond to LPS/LBP in the presence of sCD14.

Is H-4 a function blocking antibody for rat CD14 and is it formulated in amounts and in buffers appropriate for in vivo studies (i.e. no azide, no carrier protein)?

Thank you for your question. We have not tested this antibody for any functional blocking activity, nor are we aware of any publications citing use of the antibody for any blocking function. How much sample do you recommend loading on the gel for Western blot detection using CD14 (H-4): sc-515785?

We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane. A link to our complete Western blotting protocol is provided here for your reference:

works very wellThis antibody works very well by WB.We tried different conditons and optimized the loading amount and tried different dilution 1:100 to 1:800 all tried.The optimiezd dilution 1:300 is the best.

Dr.This antibody has two more non-special bands in my blot, when I detected it in the MLE-12 cells

Identifying and removing macrophageThis antibody is really good for identifying and removing macrophage as contaminating cells in isolates in conjugation with dynabeads magnetic beads. Therefore this is a must have item, will definitely be stocking up!
