caspase-3 p17 Antibody (B-4) is a mouse monoclonal IgG2a kappa light chain antibody engineered to detect the p17 subunit of caspase-3, also known as apopain, SCA-1, Yama, and CPP32. Anti-caspase-3 p17 antibody (B-4) targets amino acids 56-104 near the N-terminus of human caspase-3 p17, ensuring precise recognition of both the p17 subunit and the full-length precursor of caspase-3 in human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Caspase-3 is a vital aspartate-specific cysteine protease within the ICE subfamily of caspases, playing a crucial role in the execution phase of apoptosis, the programmed cell death process essential for development, tissue homeostasis, and the removal of damaged or unwanted cells. Structurally, caspase-3 is synthesized as an inactive proenzyme, procaspase-3, which undergoes proteolytic cleavage to form the active enzyme composed of two p17 and two p11 subunits, creating a heterotetramer essential for enzymatic activity. This structural configuration allows caspase-3 to effectively cleave key substrates such as PARP, facilitating the orderly disassembly of cellular components during apoptosis. Active caspase-3 can activate downstream caspases like caspase-6, -7, and -9, thereby amplifying the apoptotic signal and ensuring completion of the cell death process. The precise structural organization of caspase-3 is critical for function, enabling rapid response to apoptotic signals within the cytoplasm, where caspase-3 is highly expressed in vital organs including lung, spleen, heart, liver, kidney, and immune cells. caspase-3 p17 monoclonal antibody (B-4) serves as an indispensable tool for researchers investigating molecular mechanisms of apoptosis, cancer progression, neurodegenerative diseases, and other conditions where programmed cell death is key. Thank you for your question. caspase-3 p17 Antibody (B-4) is only recommended for detection of p17 subunit and full length precursor of caspase-3 of human origin by WB, IP, IF, IHC(P) and ELISA. Based on our tests we do not recommend it for use with rat samples.

Is there antibody aganist p17 only without the procaspase-3 for IHC(P)?

Can this antibody recognize Caspase 3 after being cleaved? The size is about 17kDa.

Thank you for your question. This monoclonal antibody is recommended for detection of the cleaved p17 subunit and full length precursor of caspase-3 of human origin. There is Western blot data on our website showing detection of the cleaved caspase-3 p17 in Staurosporine treated HeLa whole cell lysates.

Is there a recommended positive control for this Ab?

Thank you for your question. We recommend using the following positive controls with this caspase-3 p17 Antibody (B-4): sc-271028: Untreated (sc-2200) and Staurosporine (sc-3510) treated HeLa (sc-24698) or Jurkat (sc-24719) whole cell lysates for detection of procaspase-3 and cleaved caspase-3 p17 expression in Western blot. Untreated (sc-2239) and Kendomycin (sc-202196) treated U-937 whole cell lysates for detection of procaspase-3 and cleaved caspase-3 p17 expression in Western blot. CCRF-CEM (sc-2225), Jurkat (sc-2204), K-562 (sc-2203), and Hep G2 (sc-2227) whole cell lysates for detection of procaspase-3 expression in Western blot.

Can this antibody also recognize the p12 subunit of caspase 3?

Thank you for your question. Our caspase-3 p17 sc-271028 is specific to the precursor form of caspase-3 and the p17 subunit, and will not detect the p12 subunit.

Unfortunately, we were unable to see the p17 fragment. We're currently optimising our cell culture incubations to detect this. Otherwise, very clean WB from this Ab. Would recommend. Conditions: 40 µg protein, 1º Ab 1/200, overnight at RT, 2º Ab, 1/10,000 m-IgGκ BP-HRP (mouse IgGκ binding protein-HRP) #sc-516102, 2 hrs RT. Imaging: ECL.
