L'anticorps c-Fos (C-10) est un anticorps monoclonal de souris IgG2a κ qui détecte c-Fos, Fos B, Fra-1 et Fra-2 d'origine souris, rat et humaine par WB, IP, IF, IHC(P) et ELISA. L'anticorps c-Fos (C-10) est disponible sous forme d'anticorps anti-c-Fos non conjugué, ainsi que sous plusieurs formes conjuguées d'anticorps anti-c-Fos, incluant agarose, HRP, PE, FITC et plusieurs conjugués Alexa Fluor®.L'oncogène c-Fos a été initialement détecté dans deux isolats indépendants du virus de l'ostéosarcome murin et dans un virus de néphroblastome aviaire. L'homologue cellulaire, c-Fos, code une phosphoprotéine nucléaire qui est rapidement et transitoirement induite par divers agents et fonctionne en tant que régulateur transcriptionnel pour plusieurs gènes. Contrairement aux protéines c-Jun, qui forment des homodimères et des hétérodimères qui se lient à des éléments de réponse spécifiques de l'ADN, les protéines c-Fos ne sont actives que sous forme d'hétérodimères avec des membres de la famille des gènes Jun. Les homologues fonctionnels de c-Fos comprennent les gènes Fra-1, Fra-2 et Fos B. De plus, certains membres de la famille ATF/CREB sélectionnés peuvent former des dimères de leucine zipper avec Fos et Jun. Différents dimères présentent une spécificité et une affinité différentielles pour les sites AP-1 et CRE.

Because there is no way to know if it works on mouse tissue until we try. To avoid anti-mouse secondary, it would be nice to try anti-Fos conjugated to Alexa 488 or 594. Do you have small samples of these?

Is there a mouse-on-mouse issue, when used on mouse tissue?

Thank you for your question. There are numerous publications citing use of this mouse monoclonal c-Fos antibody on mouse tissue. Typically, unless your tissues are rich with endogenous IgG there should not be a lot of non-specific binding using a mouse monoclonal antibody on mouse tissue. However, we do offer this antibody directly conjugated, so you can avoid having to use any anti-mouse IgG secondary antibody for detection, and have the most specific result.

What is the expiration date of this ab kept at 4C?

All of our products are warrantied for one year from the date purchased. We do not provide expiration dates for our antibodies. If properly stored at 4C it can continue to be viable for several years if not longer.

Is c-Fos (C-10): sc-271243 monoclonal antibody appropriate for use in IHC with paraffin-embedded sections?

Thank you for your question. Yes, c-Fos (C-10): sc-271243 monoclonal antibody has been validated for use in IHC with paraffin-embedded sections. Rated 4 de 5 de par Worked for mouse brain tissueI tried c-Fos ab C-10, D-1, E-8 at the same time in PFA-fixed flozen mouse brain and found only C-10 worked well. I applied 1:500 ~ 1:8000 dilution in 4°C, over night for floated 40um thick-sections. 1:8000 dilution was better for me.

Rated 1 de 5 de par It is not useful for immunohistochemitryWe processed sections of the same subject with different dilutions and with a polyclonal antibody that has resulted very good for our purposes. The monoclonal c-Fos (C-10) did not give a good immunostaining. Moreover, it uncovered structures that look like blood vessels and obstruct a good appreciation of the c-Fos positive cells. It is important to indicate that such vessels were not vissible with our reference antibody.

Rated 5 de 5 de par Worked great for IFI was looking for a robust c-Fos antibody for immuno-staining in mouse brain tissue. This one worked like a champ. I used 1:300 in PBST, RT o.n. for 100um slice. For secondary, I used Invitrogen isotype specific Alexa 647 A-21241 1:300 in PBST, RT o.n. and other secondary abs yielded nonspecific signals.
