Arg2 Antibody (A-10) is a mouse monoclonal IgA kappa light chain antibody that specifically targets an epitope between amino acids 304–335 near the C-terminus of human Arginase 2 (ARG2), also known as Arginase II. ARG2 is a mitochondrial enzyme that catalyzes the hydrolysis of L-arginine into L-ornithine and urea, playing a crucial role in the urea cycle and polyamine biosynthesis. Structurally, ARG2 is a 354 amino acid protein that forms a homotrimer, with each subunit containing a binuclear manganese cluster essential for catalytic activity. The structural configuration of ARG2 is significant because the active site architecture and manganese coordination are critical for substrate binding and enzymatic function. Unlike its cytosolic counterpart Arginase I, which is predominantly expressed in the liver, ARG2 is primarily found in extrahepatic tissues such as the kidney and small intestine. The mitochondrial localization of ARG2 influences nitric oxide production by modulating L-arginine availability, thereby affecting vascular function and immune responses. Dysregulation of ARG2 has been implicated in various pathological conditions, including cardiovascular diseases, neurodegenerative disorders, and cancer. Anti-ARG2 antibody (A-10) demonstrates reactivity with ARG2 of mouse, rat, human, and additional species like canine, bovine, and porcine origin in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). ARG2 monoclonal antibody (A-10) is a valuable tool for researchers studying the structural and functional aspects of ARG2, contributing to a deeper understanding of its role in physiological and pathological processes.

Why does the image in the datasheet show a band size of 90kDa when the expected protein size is 40kDa. This antibody is suggested as a replacement for H-64 which was a rabbit monoclonal at 40kDa. So does this not detect the same protein? Thanks.

Thank you for your question. According to the literature and our data in arginase II transfected lysates, the expected molecular weight for arginase II is approximately 40 kda. Unfortunately, we do not have any reference to support a molecular weight of 90 kda in cell lines at this time. Therefore, we are going to remove that data from our website and reevaluate the antibody in different cell lines. This mouse monoclonal antibody, arginase II (A-10): sc-393496, does detect the same arginase II protein as the now discontinued rabbit polyclonal antibody.

I have two questions. First, in your data sheet, molecular weights is 40kDa. But figure of WB band says 90kDa. Is it fusion band? I wonder real band size.  Next, what secondary antibody should I use for western blotting?

Thank you for your question. According to the literature and our data in arginase II transfected lysates, the expected molecular weight for arginase II is approximately 40 kda. Unfortunately, we do not have any reference to support a molecular weight of 90 kda in cell lines at this time. Therefore, we are going to remove that data from our website and reevaluate the antibody in different cell lines. This mouse monoclonal antibody, arginase II (A-10): sc-393496, is a mouse monoclonal IgA (kappa light chain). Ideally, we would recommend using an isotype-specific secondary antibody (anti-mouse IgA) for detection. Unfortunately, we do not currently offer this type of secondary antibody for sale.

What blocking solution should I use with arginase II (A-10): sc-393496 monoclonal antibody for Western blot?
