Connexin 46 (Cx46) is a protein that forms gap junctions in the lens of the eye, facilitating intercellular communication. Direct chemical activators of Cx46 have not been well-characterized. However, several compounds may indirectly influence Cx46 function through associated pathways or cellular processes.Connexin 46 Activators and Modulators represent a diverse class of chemicals that can influence the function of Cx46, a protein critical for the formation of gap junctions in the eye lens. Direct activation of Cx46 is not well-documented in the literature. However, these compounds can indirectly influence the function of Cx46 and other connexins by modulating gap junction intercellular communication.

Compounds like retinoic acid can alter the transcription and translation of connexins, thereby potentially impacting Cx46 levels. Other chemicals, such as 18β-Glycyrrhetinic acid, quinine, carbenoxolone, heptanol, glycyrrhizin, mefloquine, and octanol, are known to inhibit gap junction communication, an effect that could extend to Cx46. Synthetic peptides Gap27 and Gap26 also inhibit various connexins and could potentially influence Cx46. On the other hand, probenecid and flufenamic acid can potentiate and modulate the function of several connexins, respectively, which could potentially affect Cx46.
