PGK1/2 Antibody (A-5) is a mouse monoclonal IgG1 kappa light chain antibody that detects PGK1/2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PGK1/2 (A-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Phosphoglycerate kinases 1 and 2 (PGK1/2) are crucial glycolytic enzymes that play a significant role in cellular energy metabolism by catalyzing the transfer of a phosphoryl group from 1,3-bisphosphoglycerate to adenosine diphosphate (ADP), resulting in the production of adenosine triphosphate (ATP) and 3-phosphoglycerate. PGK1 is secreted by tumor cells and is involved in angiogenesis, the process of new blood vessel formation, which is vital for tumor growth and metastasis. PGK1 acts as a disulfide reductase, facilitating the reduction of disulfide bonds in the serine proteinase plasmin, which triggers the release of angiostatin, a potent inhibitor of tumor blood vessel formation. The human PGK gene, located on the X chromosome, spans 23 kilobases and is interrupted by 10 introns, with mutations in this region linked to various genetic disorders, including prostate cancer and androgen insensitivity.

I am using PGK1/2 (A-5): sc-48342 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: cjMara
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 5 out of 5 by from Excellent productThe PGK1/2 Antibody (A-5): sc-48342 works well for IHC applications when using formalin fixed, paraffin-embedded tissue sections, showing staining pattern localized to the nucleus and cytoplasm. This antibody has also been cited several times for further verification.

Rated 4 out of 5 by from PGK/12 reviewI stained primate trabecular meshwork and that gave me good signal compared to just secondary Antibody control. However my positive control, mouse colon cancer tumor, I had staining with secondary only and primary and secondary. it may be that my peroxide was old. But it seems to work fine in trabecular meshwork
