p-Trk Antibody (E-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects Trk phosphorylated at Tyr 496, along with correspondingly phosphorylated Trk B and Trk C, in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). p-Trk (E-6) antibody is available in both non-conjugated and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. The Trk proto-oncogene encodes a membrane-spanning protein tyrosine kinase, Trk gp140, which is crucial for neuronal development and function, as expression is primarily restricted to neurons originating from the sensory spinal and cranial ganglia. The phosphorylation of Trk A by nerve growth factor (NGF) is a rapid and specific process that occurs even at picomolar concentrations, highlighting the physiological significance of NGF in neuronal signaling. This phosphorylation is essential for mediating the survival, differentiation, and growth of neurons, making Trk A a vital component in neurotrophic signaling pathways. In contrast, Trk B is phosphorylated by brain-derived neurotrophic factor (BDNF), while Trk C phosphorylation is induced by neurotrophin-3 (NT-3), illustrating the diverse roles of these neurotrophins in neuronal health and development.
