AICAR transformylase Activators

AICAR transformylase activators are compounds that either increase the availability of the enzyme's substrate, AICAR, or the one-carbon donor, 10-formyltetrahydrofolate.The primary action of these compounds revolves around the enhancement of the purine nucleotide biosynthesis pathway, where AICAR transformylase plays a crucial role. Substances such as AICAR, 5-CHO-THF, and 10-CHO-THF directly interact with the enzyme, acting as substrates or co-factors that the enzyme uses in its catalytic function. Their presence in higher concentrations can enhance the activity of AICAR transformylase. These compounds are fundamental to the enzymatic action of AICAR transformylase, contributing to the transformation of AICAR into FAICAR, a critical step in purine nucleotide biosynthesis.

Other compounds, including Methotrexate, Trimethoprim, Pemetrexed, Pyrimethamine, Pralatrexate, and Raltitrexed, interact with the folate pathway, causing an increase in AICAR levels. By inhibiting DHFR or other folate pathway enzymes, these compounds lead to an accumulation of dihydrofolate, which then increases the levels of AICAR. This increase in substrate availability indirectly enhances the activity of AICAR transformylase. Folic acid and folinic acid, on the other hand, can be converted into 10-formyltetrahydrofolate in the cell, therefore providing the formyl group needed for the conversion of AICAR to FAICAR. This way, these compounds can also indirectly enhance the functional activity of AICAR transformylase, leading to an overall increase in purine nucleotide biosynthesis.

Folic acid, once in the body, is converted into tetrahydrofolate, a form that can be converted into 10-formyltetrahydrofolate. The increased availability of this molecule enhances the functional activity of AICAR transformylase by providing the formyl group for the conversion of AICAR to FAICAR.
