XRN1 encodes a highly conserved 5′→3′ exoribonuclease that drives cytoplasmic mRNA turnover by degrading decapped transcripts and clearing RNA decay intermediates. It is a core component of post-transcriptional gene regulation, linking decapping and exonucleolytic decay to pathways controlling mRNA surveillance, stress granule dynamics, and innate immune sensing of aberrant or viral RNAs. Through its role in RNA quality control and remodeling of the transcriptome, XRN1 influences cellular responses to stress and infection as well as broader programs in proliferation and differentiation. Dysregulated RNA decay and impaired XRN1 activity have been associated with altered interferon signaling, neurodevelopmental phenotypes, and cancer-relevant changes in gene expression networks.
XRN1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the XRN1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the XRN1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the XRN1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish XRN1 protein expression.
This CRISPR knockout system enables efficient generation of XRN1-deficient cell models for investigation of XRN1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
