PUS1 Antibody (B-6) is a mouse monoclonal IgG1 antibody that detects PUS1 in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). PUS1, or Pseudouridine synthase 1, plays a crucial role in the post-transcriptional modification of RNA by converting uridine into pseudouridine, a modification that enhances the stability and functionality of tRNA and rRNA molecules. This modification is vital for the proper folding and function of these RNA species, which are essential for protein synthesis and cellular function. PUS1 is predominantly located in the nucleus, where PUS1 forms complexes with other proteins such as RARG and SRA1 RNA, facilitating PUS1′s role in RNA processing. PUS1 expression is notably high in brain and skeletal muscle tissues, reflecting PUS1′s importance in these metabolically active regions. Deficiencies in PUS1 are linked to myopathy with lactic acidosis and sideroblastic anemia (MLASA), a rare genetic disorder that affects oxidative phosphorylation in bone marrow and skeletal muscle, underscoring PUS1′s significance in maintaining cellular health. Human PUS1 consists of 348 amino acids and exhibits a remarkable 92% sequence homology with mouse PUS1, highlighting PUS1′s evolutionary conservation and functional importance across species.
