Flightless I Antibody (E-1) is a mouse monoclonal IgG2b antibody that detects Flightless I in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Flightless I plays a crucial role in cellular processes, particularly in actin cytoskeleton organization, which is essential for cell migration, division, and shape maintenance. Flightless I contains an Actin-binding domain sharing homology with the Gelsolin family, indicating involvement in actin filament dynamics and rearrangements. Flightless I features an N-terminal leucine-rich repeat domain facilitating protein-protein interactions, further underscoring importance in cellular signaling pathways. Flightless I predominantly localizes in the nucleus, with subsequent translocation to the cytoplasm upon serum stimulation, highlighting dynamic role in cellular responses to external stimuli. In migrating cells, Flightless I co-localizes with structures based on beta-tubulin and actin, emphasizing integral function in coordinating cytoskeletal rearrangements necessary for effective cell movement. The human FLI gene, encoding Flightless I, is located within the Smith-Magenis microdeletion region on chromosome 17 at 17p11.2, and mutations in this gene are associated with Smith-Magenis syndrome, a condition characterized by various developmental and behavioral challenges.
