DAP10 Antibody (C-5) is a mouse monoclonal IgG2a antibody that detects DAP10 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DAP10 is a transmembrane type 1 protein predominantly expressed in hematopoietic cells, and DAP10′s function is crucial for immune response regulation. DAP10 forms an activating receptor complex with NKG2D, which is essential for initiating Natural Killer and T-cell mediated cytotoxicity against tumors that express ligands such as MICA and MICB. The interaction between DAP10 and NKG2D is particularly important in the context of epithelial tumors, where both proteins, along with their ligands, are stress-inducible molecules. Notably, DAP10′s cytoplasmic region contains a binding site for the SH2 domain of the p85 subunit of phosphoinositide 3-kinase, indicating DAP10′s significant role as a signal transducer that leads to the activation of this critical signaling pathway.
