Treacle Activators

Treacle, also known as TCOF1, is a nucleolar protein essential for embryonic development, particularly in craniofacial morphogenesis. It is predominantly localized in the nucleolus, where it participates in the biogenesis and processing of ribosomal RNA (rRNA). Treacle plays a crucial role in ribosome assembly by facilitating the recruitment and assembly of ribosomal proteins onto nascent rRNA transcripts, ensuring the proper formation of ribosomal subunits. Additionally, Treacle is involved in the regulation of ribosomal DNA (rDNA) transcription and nucleolar organization, contributing to the maintenance of nucleolar integrity and function. Dysregulation of Treacle expression or function is associated with Treacher Collins syndrome (TCS), a congenital disorder characterized by craniofacial abnormalities, highlighting its importance in craniofacial development and disease pathogenesis.

The activation of Treacle involves complex regulatory mechanisms orchestrated by various cellular processes and signaling pathways. Transcriptional regulation plays a crucial role in controlling Treacle expression levels, with transcription factors and epigenetic modifiers modulating TCOF1 gene expression in a cell- and context-dependent manner. Post-translational modifications, such as phosphorylation, acetylation, and ubiquitination, regulate Treacle activity and subcellular localization, influencing its function in ribosome biogenesis and nucleolar organization.
