TMEM223 Activators

Chemical activators of TMEM223 can engage in various cellular mechanisms to enhance its activity. Forskolin serves as a potent activator of adenylyl cyclase, which in turn elevates intracellular cAMP levels. The increase in cAMP can then activate protein kinase A (PKA), a kinase that can phosphorylate a wide range of substrates including TMEM223. This phosphorylation event can lead to the functional activation of TMEM223. Similarly, PMA is known to activate protein kinase C (PKC), another kinase that plays a crucial role in phosphorylation signaling pathways. The activation of PKC can lead to the phosphorylation of TMEM223, thereby promoting its active state. Ionomycin and A23187, both calcium ionophores, can effectively increase intracellular calcium concentrations. Elevated calcium levels can activate calcium-dependent kinases, which are capable of phosphorylating and activating TMEM223.

Hydrogen peroxide functions as a reactive oxygen species that can initiate signal transduction pathways leading to the phosphorylation of proteins, including TMEM223. Anisomycin activates stress-activated protein kinases, which are also involved in phosphorylation cascades that can target TMEM223 for activation. Fingolimod interacts with sphingosine-1-phosphate receptors and can trigger downstream kinase cascades; these cascades can lead to the activation of TMEM223 via phosphorylation events. Calyculin A and Okadaic Acid both act as inhibitors of protein phosphatases, which typically reverse phosphorylation. By inhibiting these phosphatases, TMEM223 remains phosphorylated and thus in an activated state. Additionally, Bisindolylmaleimide I, although a PKC inhibitor, can alter signaling pathways such that other kinases may compensate and phosphorylate TMEM223, leading to activation. Lastly, Thapsigargin and Brefeldin A disrupt calcium homeostasis and Golgi function, respectively, which can affect signaling pathways and kinase activities, ultimately leading to the activation of TMEM223. These disruptions result in secondary effects that can enhance the phosphorylation and activation of TMEM223.
