Chemical activators of F-box and WD-40 domain protein 27 employ various cellular signaling pathways to initiate its phosphorylation and subsequent activation. Phorbol 12-myristate 13-acetate (PMA) specifically targets protein kinase C (PKC), which can directly phosphorylate F-box and WD-40 domain protein 27. Similarly, forskolin works by elevating intracellular cAMP levels, thereby activating protein kinase A (PKA), which also serves to phosphorylate F-box and WD-40 domain protein 27. Ionomycin functions by increasing intracellular calcium concentrations, which activates calcium/calmodulin-dependent protein kinases (CaMKs), and these kinases are capable of phosphorylating F-box and WD-40 domain protein 27 as well. Thapsigargin disrupts calcium homeostasis by inhibiting the sarcoplasmic/endoplasmic reticulum Ca2+ ATPase (SERCA), causing a rise in cytosolic calcium levels that similarly activate CaMKs and lead to the phosphorylation of F-box and WD-40 domain protein 27.
Other chemical activators operate through different mechanisms but achieve the same end of phosphorylating and activating F-box and WD-40 domain protein 27. Anisomycin initiates a stress response that activates stress-activated protein kinases (SAPKs), such as JNK, which in turn can phosphorylate F-box and WD-40 domain protein 27. Conversely, okadaic acid inhibits protein phosphatases, which normally dephosphorylate proteins, thereby maintaining F-box and WD-40 domain protein 27 in a phosphorylated, active state. In a somewhat similar vein, calyculin A also inhibits protein phosphatases leading to sustained phosphorylation of F-box and WD-40 domain protein 27. Meanwhile, staurosporine, at low concentrations, can activate certain kinases that may phosphorylate F-box and WD-40 domain protein 27. Epigallocatechin gallate (EGCG) inhibits some protein kinases, which may lead to the activation of alternative kinases capable of phosphorylating F-box and WD-40 domain protein 27. Sphingosine and bisindolylmaleimide I can activate kinase cascades potentially resulting in the phosphorylation of F-box and WD-40 domain protein 27. Lastly, dibutyryl-cAMP, a cAMP analog, activates PKA, further contributing to the phosphorylation landscape that regulates the activity of F-box and WD-40 domain protein 27.
