StARD8 Activators

StARD8 activators primarily function through pathways that modulate the phosphorylation state of StARD8. Compounds such as 8-Br-cAMP, Forskolin, IBMX, Rolipram, Cilostazol, H-89, Vardenafil, Sildenafil, Tadalafil, and Papaverine all work by increasing intracellular cAMP levels through various mechanisms. cAMP is a second messenger that activates protein kinase A (PKA). PKA can phosphorylate and activate StARD8, leading to an increase in its functional activity. By increasing the levels of cAMP in the cell, these compounds indirectly lead to the activation of StARD8.

On the other hand, Y-27632 and Fasudil act by inhibiting Rho-associated protein kinases (ROCK1/2). ROCK1/2 are known to phosphorylate and inactivate StARD8. Therefore, inhibition of these kinases leads to reduced phosphorylation and inactivation of StARD8, thereby enhancing its activity. Y-27632 and Fasudil are thus indirect activators of StARD8. In summary, the compounds listed either enhance the activity of StARD8 by triggering its activation through the cAMP-PKA pathway or by preventing its inactivation through the ROCK pathway.
