SF3B10 Activators

Chemical activators of SF3B10 play a significant role in the modulation of spliceosome activity through various cellular signaling pathways. Okadaic Acid and Calyculin A are known to inhibit protein phosphatases PP1 and PP2A, which typically decrease the phosphorylation levels of proteins. In the context of SF3B10, this inhibition can cause an increase in phosphorylation, thereby enhancing the protein's activity within the spliceosome complex. Similarly, Phorbol 12-myristate 13-acetate (PMA) harnesses the ability to activate protein kinase C (PKC), which is known to phosphorylate a range of proteins. The activation of PKC by PMA can lead to the phosphorylation of SF3B10, thus promoting its role in RNA splicing. Forskolin, which increases intracellular cAMP, indirectly activates protein kinase A (PKA) and can result in the subsequent phosphorylation and activation of SF3B10, impacting its splicing functions.

Ionomycin, by increasing intracellular calcium concentrations, can activate calcium-dependent kinases that may target and activate SF3B10. Staurosporine, although a broad kinase inhibitor, in specific conditions can activate kinases that potentially target SF3B10, enhancing its activity. Epigallocatechin gallate (EGCG) interacts with various cellular pathways that involve phosphorylation, which can lead to the activation of SF3B10. The cAMP analog Dibutyryl-cAMP activates PKA, possibly leading to phosphorylation and activation of SF3B10. Jaspakinolide, by stabilizing actin filaments, affects nuclear signaling pathways and dynamics, which can influence the activation state of SF3B10. Anisomycin activates stress-activated protein kinases which can phosphorylate and activate SF3B10. Cantharidin, through the inhibition of PP2A, can lead to increased phosphorylation and activation of SF3B10. Lastly, Bisindolylmaleimide I, although primarily a PKC inhibitor, can inadvertently lead to the activation of compensatory phosphorylation pathways that include SF3B10 activation, demonstrating the complex interplay between cellular signaling and protein regulation.
